BLANCHAER, M. C. Respiration of mitochondria of red and white skeletal muscle. Am. J. Physiol. 206(5) :1o15-1020. I g64.-Mitochondria of guinea pig red and white skeletal muscle were isolated in a medium containing 0.25 M sucrose, I mM ethylenediamine tetraacetate, and I y0 fraction V bovine albumin. Oxidative phosphorylation with pyruvate-malate as substrate was tightly coupled in both types of mitochondria when oxygen uptake was measured over a 2to +min period with a platinum electrode. The P/O ratios approached the theoretical value of 3. Oxygen consumption was also determined manometrically with the substrates IO mM pyruvate-1 mM malate, IO mM succinate, 6 mM reduced nicotinamide adenine dinucleotide (NADHZ), 20 mM dl-lactate, and 12-20 mM dl-a-glycerophosphate. White muscle mitochondria had a higher rate of oxygen consumption with cr-glycerophosphate than with lactate, succinate, and NADHZ. Those of red muscle were less active with cu-glycerophosphate than with the other substrates. These results indicate that an cr-glycerophosphate shuttle may couple the reactions generating NADHz in the cytoplasm of white muscle with the mitochondrial respiratory chain. The properties of the red muscle mitochondria suggest that the direct oxidation 01 mADH2 may be more important in this tissue than the a-glycerophosphate shuttle.

8-Oxoguanosine and uracil repair of nuclear and mitochondrial DNA in red and white skeletal muscle of exercise-trained old rats.

Oxoguanine DNA glycosylase (OGG1) and uracil DNA glycosylase (UDG) are two of the most important repair enzymes that are involved in the base excision repair processes to eliminate oxidative damage from mammalian DNA, which accumulates with aging. Red and white skeletal muscle fibers have very different antioxidant enzyme activities and resistance to oxidative stress. In this paper, we demonstr...

Rate of calcium binding and uptake in normal animal and failing human cardiac muscle. Membrane vesicles (relaxing system) and mitochondria.

Cardiac relaxing system (CRS) and mitochondria were isolated by a modified rapid procedure from human, rabbit, and dog. Relaxing system (sarcoplasmic reticulum) was also isolated from white and red skeletal muscles for comparative purposes. Rapid kinetic measurements were made with a dualbeam spectrophotometric assay procedure. Maximum calcium binding (absence of oxalate), expressed as nmoles c...

Skeletal Muscle-Specific Overexpression of PGC-1α Induces Fiber-Type Conversion through Enhanced Mitochondrial Respiration and Fatty Acid Oxidation in Mice and Pigs

Individual skeletal muscles in the animal body are heterogeneous, as each is comprised of different fiber types. Type I muscle fibers are rich with mitochondria, and have high oxidative metabolisms while type IIB fibers have few mitochondria and high glycolytic metabolic capacity. Peroxisome proliferator-activated receptor gamma coactivator 1α (PGC-1α), a transcriptional co-activator that regul...
